Co-amoxiclav (amoxicillin & clavulanic acid). Per Spotclav 625 625 mg FC tab Amoxicillin trihydrate 500 mg, clavulanate K 125 mg. Per 5 mL Spotclav DS 312.5 powd for oral susp Amoxicillin trihydrate 250 mg, clavulanate K 62.5 mg
Indications/Uses
Bacterial infections due to susceptible organisms eg, upper resp tract infections (including ENT) eg, tonsillitis, sinusitis, otitis media; lower resp tract infections eg, acute & chronic bronchitis, lobar & broncho pneumonia; GUT infections eg, cystitis, urethritis, pyelonephritis, pelvic infections, chancroid, gonorrhoea; other infections eg, puerperal sepsis, septic abortion, intra-abdominal sepsis; skin & soft tissue infections eg, boils, abscesses, cellulitis, animal bites, wound infection; bone & joint infections eg, osteomyelitis; dental infections eg, dentoalveolar abscess.
Dosage/Direction for Use
Spotclav 625 Adult & childn >12 yr Mild-moderate infection 1 tab bid. Severe infection Co-amoxiclav 1,000 mg bid. Moderate renal impairment (CrCl 10-30 mL/min) 1 tab every 12 hr. Spotclav DS 312.5 Childn 6-12 yr (18-40 kg) 5 mL every 8 hr or 0.4 mL/kg daily in 3 divided doses.
Administration
Should be taken with food.
Contraindications
Hypersensitivity to penicillins, cephalosporins & other β-lactamase. Patients w/ previous history of co-amoxiclav or penicillin-associated jaundice or hepatic dysfunction.
Special Precautions
Avoid use if glandular fever is suspected. Cholestatic jaundice during or shortly after use. Patients w/ GI disease. Prolonged use may result in overgrowth of non-susceptible organisms. Hepatic impairment. Avoid use during pregnancy. Spotclav 625 Not recommended in severe renal impairment (CrCl <10 mL/min).
Adverse Reactions
Diarrhoea, nausea, vomiting, epidermal necrolysis, exfoliative dermatitis, antibiotic-associated colitis (including pseudomembranous colitis), candidiasis, vasculitis.
Drug Interactions
May reduce reabsorption of oestrogen & efficiency of combined OCs. Prolonged bleeding & prothrombin time w/ anticoagulant therapy. Amoxicillin: Increased & prolonged serum & bile conc w/ probenecid. May increase incidence of allergic skin reactions w/ allopurinol.
